In-Silico Clinical Trials with NLME Models: A Pumas Demonstration on a Hepatitis B QSP Model

Cortés-Ríos et al. (CPT:PSP, 2025) recently proposed a step-by-step workflow for performing in-silico clinical trials with nonlinear mixed-effects (NLME) models. In this webinar, we demonstrate that workflow in Pumas on an 11-ODE QSP model of chronic hepatitis B (Cortés-Ríos et al., Clin Pharmacol Ther 2025).

The session covers:

  • Defining the published NLME model in Pumas
  • Global sensitivity analysis (Sobol, eFAST) to rank parameter influence
  • Structural identifiability assessment under different measurement scenarios
  • Virtual population generation via Gaussian copulas
  • Calibration of the virtual population to clinical distributions using  mixed-integer linear programming (MILP)
  • Multi-arm virtual trial simulation and visualization of outcome distributions
